PMA-mediated activation of Tg gene transcription and Tg mRNA accumulation was blocked by the protein kinase C inhibitor H-7 and by anti-protein kinase C-epsilon antibody. The effect of PMA was dependent on protein kinase C activity since a pretreatment with calphostin C prevented the increases of Tg mRNA levels by PMA. As demonstrated by nuclear run-on analysis, PMA-induced Tg gene transcription was increased by 2-fold in FRTL-5 cells. We have also shown that PMA induced a 2.5-fold increase of Tg promoter activity in FRTL-5 cells.
